I didn't know anything about FFXIV when I started reading the series (and still don't know anything that I didn't learn via osmosis by reading the series), but understood everything well enough that it didn't interfere with enjoying the story. The FFXIV elements started small and grew over time, but I thought they were explained well.
I definitely would suggest reading the works in order, since Part 1 starts in the DoS setting and introduces only a single FFXIV element, Part 2 is the 'meat' of the story that integrates both settings, Part 3 has a lot of time skips and a plot which relies heavily on the fallout from Part 2's events, and the first chapter of Part 4 takes place entirely in the FFXIV setting without even mentioning Shikako.
